Yes, you can apply from either inside or outside Australia. If applying from inside Australia, you must hold a substantive visa or bridging visa (A, B, or C).

This is a permanent visa allowing you to stay indefinitely, with a travel component valid for five years.

What happens if my EOI expires?

Your EOI remains valid for 2 years. You can update it during this period to reflect changes in your circumstances.
